چکیده

Hydrogels Kristy M. Wood and Nicholas A. Peppas The main advantage of oral protein delivery is that it improves patient compliance and comfort over other routes of administration (i.e. injection), thus leading to a more effective treatment regimen. Although oral protein delivery could benefit many individuals, maintaining the functionality of the protein and low bioavailability of the delivered drug have prevented it from becoming a successful therapy.

Oral insulin delivery systems based on complexation polymer hydrogels

The potential of complexation hydrogels composed of poly(methacrylic acid) grafted with poly(ethylene glycol) (P(MAA-g-EG)) for oral dosage forms of insulin is reviewed. The complexation hydrogels exhibit unique pH-responsive characteristics in which interpolymer complexes are formed and dissociated, respectively, in acidic and neutral/basic environments. The hydrogels are capable of highly inc...

Mucosal insulin delivery systems based on complexation polymer hydrogels: effect of particle size on insulin enteral absorption.

Insulin-loaded polymer (ILP) microparticles composed of poly(methacrylic acid) and poly(ethylene glycol), which have pH-dependent complexation and mucoadhesive properties have been thought to be potential carriers for insulin via an oral route. Nevertheless, further optimization of the polymer delivery system is required to improve clinical application. Therefore, the effect of particle size of...

Novel delivery system based on complexation hydrogels as delivery vehicles for insulin-transferrin conjugates.

A variety of approaches have been investigated to address the problems associated with oral insulin delivery, but the bioavailability of oral insulin is still low. Insulin is rapidly degraded by the enzymes in the GI tract and is not transported across the epithelial barrier easily. The oral insulin formulation developed in this work makes use of complexation hydrogels for oral delivery of insu...

Gastrointestinal transit and mucoadhesive characteristics of complexation hydrogels in rats.

The aim of this study was to investigate the gastrointestinal (GI) transit and mucoadhesive properties of complexation hydrogels, poly(methacrylic acid-grafted-ethylene glycol). The fluorescent labeled complexation hydrogels containing different molar ratios of methacrylic acid/ethylene glycol and different particle diameters were synthesized by free radical solution polymerization. The GI tran...
